Purification of Hemolysin Produced by Vibrio cholerae non-O1 Isolated from Seawater

Abstract
Vibrio cholerae non-O1 strain producing hemolysin and protease was isolated from seawater. The isolate showed a 307 bp band corresponding to cholera toxin gene (ctx) by polymerase chain reaction (PCR). but could not produce cholera toxin like V. cholerae O1. V. cholerae non-O1 isolate showed the highest hemolytic activity at late log phase during growth. Hemolysin was fractionated by FPLC with phenyl sepharose column (HP 16/10) and partially purified with 2,631 HU/mg. The antiserum against partially purified hemolysin was prepared using a male rabbit. Hemolysin neutralization score was 2,460, and significant inhibition on hemolytic activity was observed on sheep blood agar plate. These results suggest that the antiserum prepared with highly purified hemolysin could be used for rapid diagnosis of pathogenic vibrio infection and inhibit vibriosis.
